Re: Pick your one guitar, one pedal and one amp...
I actually use a one guitar, one pedal, one amp setup in one of my bands.
Partscaster Esquire into a pedal that was sold as a whooly mammoth clone but isn't at all into a Laney LH50R (with a Kustom DE610 bass cab as I don't own a guitar cab). Sounds awesome. The pedal is similar to my Russian Muff but much tighter at the bottom end. I use a tuner as well, though.

What I tend to play with all the time:
Thinline Telecaster > Sarno Earth Drive > '66 Princeton Reverb
Thinline Telecaster > Sarno Earth Drive > '66 Princeton Reverb
The cool thing about fretless is you can hit a note...and then renegotiate.

Re: Pick your one guitar, one pedal and one amp...
Maybe it's that I haven't played this guitar in a while because I broke a bridge saddle, but today I'm surprised that I feel like saying...
my p90ish guitar in my personal baritone tuning (AEBEAD)
•Charvel Skatecaster with a surf90 in the neck and a mean90 in the bridge, also a floyd rose ( in so many ways, but I've come to really like it)
almost said space echo, but I think without ever playing a real one I'm still gonna say
•Echorec
80--100w danelectro amp I have that sounds massive through a bass cab. I have an older Mesa Diesel 2x15 cab that someone took off the tolex, stained and oiled the plywood, and installed some kind of OEM Eminence 100w speakers. The previous owner said they were prototypes made for Ashdown. Idk, but this set up is airy and chimey with powerful low mids. With guitar it's almost too tight and powerful. And with bass it's a little too mushy and not enough volume on it's own with a band. But it sounds great with either, and with a baritone tuning and p90s it's jazzy to sweet to mean and barky. With distortion or fuzz it sounds so good it makes me want to learn metal.
•Danelectro DS-100 head + 2x15 cab
More traditionally, I'd say maybe
•a strat, and let's just say only the middle pickup
•digital reverb, maybe by Mr. Black
•a nice Fender Pro Reverb
